摘 要:
切取 8 d 苗龄的尾叶桉幼苗下胚轴, 培养 7 d 后愈伤组织的诱导率达 100%。8 周后, 愈伤组织分化成褐色、白色和浅 绿色 3 种类型。其中只有浅绿色愈伤组织能分化出不定芽, 分化率达 57%。浅绿色愈伤组织的过氧化物酶(POD)、超氧化 物歧化酶(SOD)、过氧化氢酶(CAT)和多酚氧化酶(PPO)的活性最高。除 SOD 外, 浅绿色愈伤组织的 POD、CAT 和 PPO 活 性与其他两类愈伤组织相比具有显著性差异(P<0.05)。H2O2 和丙二醛含量则是褐色愈伤组织的最高。关键词:尾叶桉; 愈伤组织; 过氧化物酶; 过氧化氢酶

Abstract:
Hypocotyls excised from 8 day-old seedlings could dedifferentiate and form callus at 100% when they were incubated on a modified MS (SPCa) medium for 7 days. Fifty-six days later, the callus differentiated into 3 types, including brown callus, white callus and reseda callus. Only could the reseda callus formed adventitious bud at 57% when these calli inoculated with the bud induction medium. The activities of POD, SOD, CAT and PPO of reseda callus were the highest. Compared with the enzyme activity of other types of callus, the POD, CAT and PPO activities of reseda callus showed significantly difference at 0.05 level, except for the activity of SOD. The content of H2O2 and malonaldehyde of brown callus was the highest.Key words: Eucalyptus urophylla; callus; peroxidase; catalase
